Keeping the original NP420 (NP = New Process) 4spd is an unrealistic goal when putting a diesel in something as big as a W500.
Prepare to spend some significant $$$.
The most practical conversion would be a Cummins 6BT coupled to a NV5600 6spd. Gearing could probably be left alone in that case.
There are a large number of other possibilities like the 4-53 Detroit Diesel I keep seeing on Ebay for $2999.
How big is the engine compartment on your W500? and just how much are you willing to spend? How much power do you want, how fast do you want to go, what size tires do you have or can you put on, what's your axle ratio?
