That hole is there to let the air out so it doesn't get air-locked. It doesn't make any difference on pressure, since pressure can get from the block to the radiator just fine through the lower hose. Pressure is determined solely by the radiator cap. Of course with the thermostat missing, it might have never gotten hot enough to build up any pressure.
