Which type of split ring are you refering to? If it's a PW, then no it's not a problem to reinflat a flat tire. Once the tire has been mounted and inflated the tire bead holds the ring against the rim groove. If you don't have the air tool that lock on the stem, then step to the side of the tire when filling as a precaution.
