Around about 1990, I went through a string of fuel delivery issues on the '55 Pontiac I had resurrected from being a parts car multiple times over.

One of these issues was with a "strip" of silicone sealant that had wedged itself inside the fuel pickup line, inside the tank. So it would run without issue via separate tank, via the stock fuel line, excepting what went inside the tank. I have no idea where that silicone came from. It appeared to be like a large amount of squeeze out along an over-sealed valve cover or oil pan rail had been peeled off and placed in the tank, then subsequently it was pulled into the fuel line, out of sight. Not a full blockage most of the time but the engine would starve for fuel after extended cruising speeds and once in a while it would be worse (perhaps the silicone kinked inside the fuel line or something to block it more than usual).

So yes, isolating the tank pickup tube from the rest of the system could be a necessity in tracking down this issue.
