That is the most direct way of getting the ratio, but if you don't feel like jacking up both sides of the vehicle, then you count the ratio between the driveshaft and wheel, and then multiply this ratio by 2 to get the actual differential ratio.
Also, many times if you jack up both tires and turn the driveshaft, one tire will have more resistance to rotating than the other, so only one will end up turning anyway. At least that's been my experience.
