The school I went through had two programs. The first, intended to produce road-ready long-haul truckers, was 4 weeks, 5 days a week, full time. The other, that I took, was called a Refresher course, and it was 3 weekend days, 7-3:30 one-on-one with the instructor. It was kind of like a fire hose of information, but the instructor was really good.
I'd say about 1/2 of what you need to learn is driving skills (particularly backing skills, which aren't trivial with a long trailer). The other 1/2 is lots and lots of details of regulations, the pre-trip inspection walkaround, air brake tests, etc. Lots of specific phrases to remember that the DMV wants to hear during your test. I haven't memorized that much stuff since high school!
