'55 Dodge 331 hemi/5speed?

I am dealing with a fellow who has a 331 Dodge hemi with a 5 speed trans from a '55 Dodge big truck. He is checking on the 5 speed being overdrive, not sure. From the pictures, it appears to have a p.t.o. opening on both sides. Looks massive, too.
Question is, would I be better off putting in my 318 poly/np435 combo or the 331 setup? Sorry for the long question.
