all tires on standard split rims must us a tube/and flap as the wheel is not air tight
you should check with the tire mfgr and see what tube/flap are recommended. some tubeless tires have internal ridges which will not work with tubes.
always get the straight story from the mfgr of the product
example michelin tech books list the acceptable rim sizes (and TYPES) along with the correct tube and flap
yes heavy equipment multi-piece wheels are tubeless but they use o-rings to make the seal between the pieces.
