Do some diagnostics on the engine. Use a vacuum gauge. They can tell you a lot. You may have a burnt valve or a bad head gasket near that one cylinder.

Also use a compression tester. Get the engine to operating temperature then pull all plugs and test all cylinders. As soon as you finish that squirt oil into each cylinder and check the compression again. If the low pressure cylinder gains a fair amount of compression with oil in it verses dry that indicates a worn cylinder and rings.
