Put it in neutral and jack up one side of the diff you want to check. Then see if you can turn the wheel by hand. If it is a locker (such as a Detroit No-spin), you shouldn't be able to turn it at all. If it is a limited slip (or posi or whatever you prefer to call it), it will be difficult to turn. If it just spins freely, turning the driveshaft it is an open diff (or roached limited slip). If you are checking the front be sure to lock your hubs if so equipped.
